Bill Kirchen did his '66 Tele that way (or actually, IIRC, had Dan Erlewine do it), not for tone, but to facilitate repeated break-down for flying as carry - on (detune a bit, capo to hold the strings to the head, a minute or so with a cordless screwdriver, and Bob's yer uncle - briefcase guitar with no wood strippage).
If you're not in the habit of removing the neck often, you can greatly enhance the grip by reaming the neck holes just enough that the screw threads slide through, thus only biting in the body wood; pulls the neck very tight into the joint.
